Drug Description:

Norethisterone Acetate is used to treat women with abnormal bleeding from the uterus. It is also used to treat women who have stopped having menstrual periods for several months due to amenorrhea, but who are not pregnant or going through menopause. Norethisterone Acetate is also used to treat a condition called endometriosis, in which tissue that normally lines the inside of the uterus is found outside the uterus in the abdomen/pelvic area, causing painful/irregular periods. Norethisterone Acetate is a type of female hormone called progestin, which is similar like the hormone progesterone that your body makes naturally. Norethisterone Acetate works by stopping the normal growth of the uterus lining during the menstrual cycle and signaling hormone changes in the uterus to restore normal menstrual periods. This drug can be used as monotherapy or in combination with other medicines. Norethisterone Acetate is administered through oral route by way of uncoated and film-coated tablets.

Mechanism of Action:

Progestins like Norethisterone Acetate exert their effects on target cells via binding to progesterone receptors that result in downstream changes to target genes. Target cells are found in the reproductive tract, breast, pituitary, hypothalamus, skeletal tissue, and central nervous system. Contraceptive efficacy is derived mainly from changes to the cervical mucus, wherein Norethisterone Acetate increases the cell content and viscosity of the mucous to impede sperm transport and migration. Norethisterone Acetate also induces a variety of changes to the endometrium that make it inhospitable for implantation. Working via a negative feedback loop, Norethisterone Acetate also acts on both the hypothalamus and anterior pituitary to suppress the release of follicle-stimulating hormone (FSH) and luteinizing hormone (LH) from the anterior pituitary. Suppression of these hormones prevents follicular development, ovulation, and corpus luteum development.
